Which biological patterns have been demonstrated to be Turing patterns?

A couple. (As usual, it depends.)Instead of only considering the classic case of reacting and diffusing chemicals, as Turing proposed in his 1952 paper, “local activation and long-range inhibition” is a general principle that may be present in various forms throughout nature. Other systems, biological and otherwise, display Turing-like patterns.Ripples of sand (or snow), patterned by the wind. Zebra stripes, patterned by … ? Note one characteristic of Turing patterns: growing stripes split rather than exceed a critical width.Of course, a lot of these patterns are probably not purely the result of Turing patterns. They are layered with… cell migration, differentiation, proliferation, positional information gradients, planar cell polarity, clock-wavefront, lateral inhibition… and other mechanisms of morphogenesis that get less attention from non-biologists. As for biologists, they mostly lost interest in Turing in the 80s after no one found any Turing systems, and Drosophila studies established that they weren’t necessary to create periodic patterns anyway.But say we’re looking for Turing mechanisms. Mere similarity of patterns is not evidence; experiments are necessary. (No, not just computer simulations.) In particular, we want to know the mechanisms responsible for local activation and long-range inhibition, and test them in model organisms.So, unfortunately, it’s not feasible to prove anything in zebras. Let’s try the next closest thing: zebra-fish!Warning: Extremely long answer ahead, because “proving” things in biology is hard. But lots of pretty pictures! Stick around!tl;dr: The most evidence is in zebrafish, and the bulk of this answer reviews those results. At the end, I added a paper each for a couple different possible Turing systems, like mice (digits, mouth bumps, hair follicles) and lizard scales.Stripe patterning in zebrafishZebrafish normally have horizontal black and yellow stripes, created by the accumulation of pigment cells called melanophores and xanthophores. But various mutants display different Turing-like patterns, suggesting that this is tantamount to tweaking the parameters of a Turing-like system (Watanabe & Kondo, 2011):Experimentally removing a patch of pigment cells with a laser allows the remaining cells to repopulate the patch (A), matching what would be predicted by Turing pattern rules (B). (Kondo & Miura, 2010)Local activation: xanthophores repel melanophores that they come in contact with. As a result, they increase the local concentration of xanthophores; i.e. local activation. The repulsive behavior is lost in cells from jaguar mutant fish, which lose hard boundaries between stripes (Inaba et al. 2012).Long-range inhibition: Melanophores are likely to die if too far away from a xanthophore. This survival signal is mediated by Delta/Notch signaling and can be modulated by increasing or decreasing Delta and Notch. Notably, Delta and Notch are membrane-bound proteins — they do not diffuse, so they require direct contact between cells. Remarkably, melanophores extend long protrusions that span stripes; these could be sites of Delta/Notch signaling (Cohen et al. 2010) and limit the maximum thickness of the stripe (Hamada et al. 2014).This interacting model of two cellular components is sufficient for a Turing-like system, albeit one carried out not by diffusing molecules, but cell-cell contacts (Kondo & Miura 2010):Because it’s biology, it’s never that simple. A third pigment cell type (silvery iridophores) is necessary for understanding how the pattern develops. Live imaging and lineage tracing in zebrafish embryos demonstrated that stripes are actually initiated by iridophores, which form a prepattern that is filled in by melanophores. That is to say, the Turing-like system between xanthophores and melanophores isn’t actually needed to pattern stripes during development!A developmental timeline that doesn’t require the aforementioned xanthophore/melanophore Turing pattern (Singh et al. 2014)Disrupting this iridophore prepattern distrupts stripe formation, allowing for some classic Turing patterns to emerge. choker mutant adults (Frohnhöfer et al, 2013):Together, positive and negative interactions between all three cell types are needed to create and stabilize zebrafish stripes. Does this still count as a Turing pattern?(Text for diagram)S-iridophores attract melanophores in high numbers, induce their aggregation into the prospective stripe area (positive long-range effect) (1) and are to some extent able to exclude them from the interstripe (negative short-range effect) (2). Xanthophores have a minor effect on melanophore aggregation (positive long-range effect) (3), but keep melanophores out from interstripe regions resolutely (negative short-range effect) (4). Interactions between iridophores and xanthophores are required to confine the shape of interstripes (5,6).And… everything else (lightning round!)Hair follicle spacing in mice (Sick et al. 2006)Historical mention to this paper since it was one of the first to bring Turing back into biology since its abandonment (The Turing Model Comes of Molecular Age). By comparison, though, it doesn’t do as good a job at demonstrating that reaction-diffusion, rather than mechanical or lateral inhibition mechanisms, are central. They didn’t even experimentally perturb Wnt, just Dkk! :(Below: Transgenic expression of inhibitor Dkk2 increases hair spacing at medium expression and greatly disrupts hair production at strong expression level.Local activation: WntLong-range inhibition: DkkPalatal ridges (rugae) in mice, you know, the things on the roof of your mouth (Economou et al., 2012)Below: If you cut out a stripe (r2), its neighbor (r3) will react by branching, as though part of a Turing pattern (shown experimentally in f, g simulated in h, i). Lateral inhibition would instead predict the formation of a new ridge in the dashed area, because “new expression contiguous with existing expression is forbidden” (d).Rugae (palatal ridges) appear to be controlled by FGF and Hedgehog. Experimentally perturbing these signaling pathways with FGF inhibition (c) or Shh inhibition (f) ex vivo caused thinning and broadening of stripes, respectively:Local activation: FGFLong-range inhibition: ShhAlso likely important: BMP4, Wise, WntDigits in mice (Sheth et al. 2012, Raspopovic et al. 2014)This one is nice because unlike rugae, everyone knows what fingers are! Sheth and colleagues reported that the dosage of certain Hox genes appeared to tune the periodicity of a Turing pattern in mouse paws:From microarray and whole-mount ISH data, Raspopovic and colleagues suggested that digit formation is partly controlled by a three-component Turing-like system composed of Bmp, Sox9, and Wnt:What I thought they did really nicely was not only create computer simulations (black and red) but also perform limb-explant studies with Bmp and Wnt inhibition (using inhibitors LDN and IWP2), demonstrating that these molecules control finger size and spacing.The implications for the evolution of digits are interesting, too — Sheth and co-authors draw comparisons to fishes and suggest that changing the number of distal Hox genes (e.g. by duplication) could have influenced the evolution of fin ray vs. digit number.Left/right patterning of the zebrafish embryo, AKA, isn’t this just a gradient? (Muller et al. 2012)It’s not a fancy pattern, but I liked that they actually used fluorescence photobleaching to measure the different diffusion/clearance rates of the molecules Nodal and Lefty in vivo. This kind of experimental evidence is hard to come by. A classic reaction-diffusion pair.
