I can’t say I know very much about this topic but i’ll try to answer to the best of my ability.Neural plasticity relates to altered dendritic function, synaptic modulation, long-term potentiation, axonal sprouting, neurite extension and growth alongside neurgenesis and synaptogenisis.Clinical usage:VPA is used to prevent seizures and epilepsy. It is also used in bipolar disorder and to prevent migranes. It has a host of side effects that range from mild to very dangerous.Mode of action:The mechanism through which valproate (VPA, also known as valproic acid) is not fully understood. It seems to prevent seizures by reducing PIP3 which is used extensively in signalling within the cell. Its also acts as an anticonvulsant by blocking sodium channels in the brain and increasing levels of neurotransmitter GABA which has inhibitory effects (compared to other neurotransmitters such as glutamate that have excitatory effects).Plasticity:According to Zhang et al., (2003) sodium valproate has shown to impair synaptic plasticity (investigated by long-term potentiation (LTP) and long-term depression (LTD) in hippocampus of rats). Impairment could be due to inhanced inhibitory transmission carried out by GABA receptors. Another study based on rat amygdala slices by Gean et al., (1994) shows valproic acid suppresses synaptic plasticity by reducing excitatory post synaptic potential of NMDA receptors (which are cruicial for plasticity).An article in the new scientist shows that VPA can increase pitch perfection in participants. Since VPA is a mood stabalizer, its role as neuroprotector has been hinted at. VPA seems to activate MAP kinases which play a role in mediating long-term neuroplastic events by effecting neurotropic factors such as Nerve Growth Factor (NGF) and Brain derived neurotrophic factor (BDNF) to promote neurite growth (Manji et al., 2000) & (Yuan et al., 2001)VPA also seems to induce alpha-synuclein (which has neuroprotective effects) by inhibition of histone deacetylase (histone deacetylase is an enzyme that allows histones to wrap around the DNA more tightly, and DNA expression is regulated by acetylation and de-acetylation) alongside preventing glutamate (an excitatory neurotransmitter) toxicity (Chuang & Leng, 2006).A very good review about the effects of valproate is carried out by Johannessen et al., (2003).Summary:VPA has clinical uses especially in epilepsy and acts as a mood stabalizer in bipolar disorder. It has many side effects.Some studies show VPA to have a detrimental effect on synaptic plasticity according to rat model.Other more recent studies show VPA to have many neurotropic effects that include changes in DNA expression, effects on MAP kinases to increase neuroplastic events leading to neurite growth and increased in pitch perfection in participants.Interesting question.
