Has there been recent studies (or conclusive results) of the connection between magnesium deficiency and Alzheimer disease?

Alzheimer's disease (AD): Could the Amyloid Hypothesis Be Wrong?The notion of abnormally folded protein(s) accumulating within neurons and destroying them aka the Amyloid - Wikipedia cascade hypothesis (Biochemistry of Alzheimer's disease - Wikipedia) has been the linchpin of AD research since the 1980s. Problem is therapies that apparently effectively removed amyloid protein from the brain led to hardly any clinical improvement in treated AD patients. Such results have brought the hypothesis itself into question (1).As the amyloid hypothesis-based AD clinical trial failures began accumulating in recent years, one group of researchers appears to have chosen to double down on the amyloid idea, arguing focus should shift towards early treatment and prevention (see below from 2).'David Knopman, a professor of neurology at the Mayo Clinic in Rochester, Minnesota...“There is mounting evidence -- of which this is another piece -- that removing amyloid once people have established dementia is closing the barn door after the cows have left,”'Such thinking also drove the impetus for the FDA to change focus towards early-stage disease (3).Meantime the problem is other plausible hypotheses don't seem to have broad acceptance within the field. Thus AD research is currently in the throes of an identity crisis of sorts.The ensuing vacuum has created fertile soil for alternative hypotheses to gain currency. Thus, some are following the trail of oxidative stress in neurons while others have chosen to focus on another protein, Tau protein - Wikipedia, also known to accumulate abnormally in AD brains (1). Link with low magnesium levels however seems fairly low on the totem pole.AD & Magnesium: Nebulous ConnectionData from human studies on a possible link between magnesium and Alzheimer's disease (AD) are sparse.A 2014 study on 1406 individuals followed for 8 years reported high magnesium intake to be related to lower risk for mild cognitive impairment (4).A 2012 study on 17-year follow-up of 1081 participants aged 60 years or older found higher the self-reported dietary intake of magnesium, lower the risk of all-cause dementia (5).A 2007 study reported low brain magnesium levels, especially in hippocampus, of AD patients (6).Such studies suggest a measure of correlation but cannot shed light on the issue of causality.OTOH, much of the evidence for a role for magnesium in Alzheimer's disease (AD) comes from animal models. However, such models recapitulate the human disease poorly or not at all, meaning not much insight to be gained on human AD.That leaves human epidemiological studies.A 2015 systematic review (the one referenced in the question, 7) analyzed whether a person's magnesium status contributed to their susceptibility for Alzheimer's (AD). Authors screened 192 studies, short-listed 13 (559 Alzheimer's, 381 healthy controls, 126 medical controls) and found that AD patients hadSignificantly lower levels of magnesium in cerebrospinal fluid (CSF) and hair compared to healthy controls.Significantly lower levels of magnesium in plasma and red blood cells compared to medical controls (these patients had diseases and/or clinical symptoms similar to those with AD).Role for magnesium in AD is speculative and ambiguous at best (8).One idea is that by modulating calcium influx through it, neuronal magnesium plays an essential role in the functioning of the NMDA receptor - Wikipedia, which is important in learning and memory.A second one hypothesizes magnesium influences neuronal oxidative stress.However, it's unclear how either notion leads to the actual AD pathology in the form of Neurofibrillary tangle - Wikipedia within neurons followed by neuronal death.Bibliography1.
