The most commonly used method in clinical practice is the "closed technique", which is considered as the standard blood collection techniques.In this method, the veins of the severed umbilical cord are cannulated and through the help of the needle the cord blood is collected into the bag. On average, the technician collects approx 75 ml of cord blood.This collected cord blood is completely depleted from red blood cells and then it is cryopreserved. Followed by storage of the same in the cord blood bank which is then further used for the transplantation.It ensures the high rate of stem cell recovery.All the patients before the procedure are required to sign informed consent forms prior to the collection of cord blood.All mothers are tested for infectious diseases as is typically done with a blood donor. For e.g Triple marker.Furthermore, the collected samples are tested for microbial sterility using an automated system.Cord blood samples are obtained under the presence and consent of the patient’s caregiver.There are three methods of collection in common use:1. One is to hang a blood bag lower than the level of the mother and let gravity draw blood down into the tube getting collected in the bag. It is mostly used in most countries worldwide, because it has fewer steps, and therefore the opportunities for mistakes are also less with less contamination.2. The second method is to draw blood actively. The draw can be done with a standard syringe or with a bulb in the tubing of the blood bag which creates suction. Studies have shown that when blood is drawn actively it will collect a larger volume with a faster speed.3. Some banks collect cord blood “outside the uterus" called as ‘ex Utero’. In this, until the placenta is delivered out trained technician needs to wait and then takes it into another room and puts it on a high shelf so that all of the blood in the umbilical cord and placenta can be drained out.In major cases, the collections are made after delivery of the baby and ligation of the cord is done prior to the expulsion of the placenta.
