*Introduction & Route of infection;------------------------------------------------"Prion" is the short name of "proteinacious infectious particle" it's a special form of mis-folded protein,The normal form is found on the surface of cells of central nervous system of humans and animals e.g. Cows,Some say that this prion protein has protective role in nerve cells and helps them respond to oxygen deficiency.It's not a virus or bacteria ,,, this agent is extraordinarily stable; neither irradiation with ultraviolet light, nor treatment with formaldehyde, nor boiling in caustic soda has any impact on its infectivity.Transmission is mainly by consumption of meat from infected animals .- Infection in animals is called: bovine spongiform encephalopathy [mad cow disease].- Infection in humans has many variants called: variant Creutzfeldt-Jakob diseases (vCJD).The first breakthrough was recorded in Britain in 1980s when mad cow disease, began to spread like wildfire through British cattle.By the 1990s it was clear that consumption of meat from infected animals was causing people to come down with variant Creutzfeldt-Jakob disease (vCJD). This and a dozen other similar diseases are designated transmissible spongiform encephalopathies (TSEs), because the brain of an infected organism [human or cattle] takes on the appearance of a sponge.Prevention of this infection by vaccines is unlikely because normal and infectious prions have the same primary structure, the human Immune system is unable to recognise infective prions as alien and so there's no immune response against them.Also it's unlikely to detect it by immunological and serological tests e.g.PCR , because there's niether nucleic acids nor Antibodies against it, to be detected.-----------------------------------------------------------------------------------------------------*How prions attack humans :------------------------------------------After ingestion of these misfolded Prions they can't be digested ,, Attempts to digest it with specific enzymes such as proteases or ribonucleases have failed.The clusters of infectous prions in GIT, break apart eventually and the infectious fragments migrate along peripheral nerves of GIT to the brain where they induce nerve cell destruction.These mis-folded proteins do not multiply (like any other virus or bacteria) in the host organism that they infect. Instead, they affect the brain structure by acting as a template, inducing proteins with normal folding to convert to a new abnormal prion misfolded form.These newly formed mis-folded proteins, in turn, act as further templates for the conversion of more normal proteins, leading to an exponential accumulation of prions in the tissue of the central nervous system. These abnormally folded proteins form plaques which are thought to cause "entanglement" of neurofibrils and interfere with synapse function. The nerve cells are eventually damaged and lost, which causes tiny vacuoles to form in the brain. These give the brain a sponge-like appearance under the microscope, hence the term spongiform disease arose.This leads to the symptoms of prion disease, which include:impaired brain function;changes in personality,memory and behavior changes & intellectual decline,movement abnormalities, particularly ataxia.These symptoms usually develop during adulthood and worsen over time, eventually causing death within several years or even a few months.
